ABSTRACT

Objective:To predict the potential molecular mechanism of Yangxue Antai Fang in treating prethrombolic state of recurrent spontaneous abortion (RSA-PTS). Method:The chemical constituents and drug targets of Yangxue Antai Fang were collected by Integrated Pharmacology-based Research Platform of Traditional Chinese Medicine (TCMIP V2.0). RSA-PTS disease target information was collected by TCMIP V2.0 and improved by Online Mendelian Inheritance in Man (OMIM) database. The interaction of these targets was analyzed and key target network was constructed. Gene ontology (GO) function and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis were further performed. Finally, Cytoscape 3.5.1 was used to build up a multidimensional network of TCM-ingredient-target-pathway. The levels of absorption, distribution, metabolism, excretion and toxicity (ADMET) of the main components in the network were analyzed. Result:A total of 310 chemical constituents and 975 targets were collected from 8 TCMs in Yangxue Antai Fang. A total of 143 targets of RSA-PTS were obtained. A total of 243 core targets were obtained by the interrelationship analysis of drug and disease targets. The analysis of the top 100 core targets showed that these targets might participate in treating RSA-PTS by affecting biological processes related to thrombosis, such as blood coagulation, platelet activation, positive regulation of angiogenesis and so on. Pathway analysis showed that these targets were mainly concentrated in complement and coagulation cascades, platelet activation, estrogen signaling pathway, thyroid hormone signaling pathway, etc. Multidimensional network analysis in combination with ADMET level showed that 14 components (leonurine, paeonol, vanillin, and so on) may play a therapeutic role in RSA-PTS by affecting coagulation factors Ⅱ (F2), plasminogen (PLG) and estrogen receptor 1 (ESR1) proteins involved in complement and coagulation cascades, platelet activation, thyroid hormone signaling pathway and others. Conclusion:The main chemical constituents in Yangxue Antai Fang may improve RSA-PTS by regulating complement and coagulation cascades, blood coagulation, platelet activation and other biological processes.

ABSTRACT

The mother can deliver nutrients to the fetus only through the placenta during pregnancy. Folic acid plays an important role in placenta development and fetal growth, and is involved in cell division, embryogenesis and fetal growth. The ability of transplacental folic acid is one of the limiting steps of folate application. Folic acid and its active forms arrive at the fetus by three transporters: folic acid receptor, reduced folic acid carrier and proton coupled folic acid transporter. The expression and transport capacity of transporters were changed significantly with pregnancy complications such as hypertension, fetal growth restriction and premature delivery. Antiepileptic drugs, progesterone, antihypertensive drugs, drug abuse, and heavy metals can alter the transport of folic acid and across the placental barrier of other drugs by regulating the placental folate transporter.

ABSTRACT

OBJECTIVE: To research the cognition, attitude and practice, the present situation and expectation obstetric medical staff on pharmaceutical care, provide the basis for establishing pharmaceutical service pathway for pregnant and delivery women. METHODS: Design questionnaire refer to literature and based on our actual situation, questions include individual choice and multiple choice, one score for every choice. The 202 questionnaires are sent to the 6 inpatient and 2 outpatient wards, the data were analyzed by SPSS20.0 statistical software. RESULTS: Response rate is 100%, and the effective rate is 99.5%. Medical staffs welcome pharmaceutical service, the cognitive score is high, at the same time, they think there are some difficulties need to be solved, which include the pharmacists' knowledge, skills and attitude the leaders, etc. The service forms include printed drug and disease information, pharmacy information query machine in hospital. The service contents mainly include counseling and guidance, adverse drug reaction monitoring, drug related problem analysis, pharmaceutical education and individualized dosage project. CONCLUSION: Obstetric medical staff provides important policy support on pharmaceutical service pathway including the opportune moment, forms and contents.

ABSTRACT

Fifteen compounds were obtained from the twigs and leaves of Caesalpinia minax. Their structures were identified as apigenin (1), 5,7,3',4'- tetrahydroxy-3-methoxyflavone (2), luteolin-5, 3 '-dimethyl-ether (3), thevetiaflavon (4), apigenin-7-O-beta-D-glucuronide (5), bonducellin (6), 7-hydroxy-3-( 4-hydroxybenzylidene )-chroman-4-one (7), 3-deoxysappanchalcone (8), 5-acetonyl-7-hydroxy-2-methyl chromone (9), 4-(trans)-acetyl-3,6,8-trihydroxy-3-methyl-dihydronaphthalenone (10), 4-(cis)-acetyl-3,6,8-trihydroxy-3-methyl-dihydronaphthalenone (11), vanillic acid (12), omega-hydroxypropioquaiacone (13), syringaresinol (14) and uracil (15). All compounds were isolated from C. minax for the first time. Compounds 1-14 were phenolic compounds and compounds 1-5, 9-13 and 15 were isolated from the genus Caesalpinia for the first time.

ABSTRACT

OBJECTIVE: To evaluate the efficacy of Marvelon combined with metformin in the treatment of polycystic ovary syndrome(PCOS). METHODS: Both prospective and retrospective clinical studies on the efficacy of Marvelon combined with metformin were searched in several databases of medicine and pharmacy, and the data was analyzed by evidence-based literature analysis method. RESULTS: There were nine randomized controlled trials(RCTs) in Chinese on Marvelon combined with metformin for the treatment of PCOS. Marvelon combined with metformin could improve hormone levels, promote ovulation and pregnancy rate markedly. CONCLUSION: It has been demonstrated in the researches that Marvelon combined with metformin is more effective for polycystic ovary syndrome(PCOS), however, further randomized, double-blind studies with larger sample size are needed to prove the efficacy and safety of Marvelon combined with metformin.

ABSTRACT

<p><b>OBJECTIVE</b>A stable primary breast cancer model in liver-specific insulin-like growth factor 1 (IGF-1) deficient (LID) mice and control mice was established. To screen apoptosis related genes expression in different serum IGF-1 levels by gene chip and flow cytometry.</p><p><b>METHODS</b>The LID mice and control mice were used. Induction of breast cancer was achieved by using the 7,12-dimethylbenz(a) anthracene. Ginsenoside Rg3 was used to interfering therapy treatment. The incidence of breast cancer in every group was compared, and expression of apoptosis associated genes was detected by gene chip and flow cytometry.</p><p><b>RESULTS</b>The incidence of tumor in none ginsenoside Rg3 injected control mice was 66.7%. The incidence of tumor in ginsenoside Rg3 injected LID mice was 12.0% which was significantly lower than any other group (P < 0.05). The apoptosis percentage in none ginsenoside Rg3 injected control mice was (2.7 +/- 0.7)%. The apoptosis percentage in ginsenoside Rg3 injected LID mice was (14.0 +/- 1.7)%. The results of gene chip indicated that in contrast to LID mice, LTA, LTB, TNF-alpha, TRAIL, TRANCE, BLK, BOK, CASP8, TRAF5, and APAF1 genes were down-regulated, and LTBR, TRAF4 genes were up-regulated in the breast cancer tissues of control mice. Application of ginsenoside Rg3 therapy could change the expression of these genes.</p><p><b>CONCLUSIONS</b>Circulating IGF-1 levels play a role in the onset and development of breast cancer. Degrade serum IGF-1 level is able to promote apoptosis by affecting the expression of a series of apoptosis related genes consequently inhibit the growth of breast cancer. There was a synergistic effect with the application of ginsenoside Rg3.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To summarize our hospital's experience in the diagnosis and treatment of Candida infection in patients with acute necrotizing pancreatitis (ANP).</p><p><b>METHODS</b>Seventy-eight cases with ANP were reviewed. There were diagnoses either by operative finding or by CT scanning. Sixty-two cases received prophylactic antibiotic treatment, other sixteen did not. For cultivation of Candida, blood, urine, stool, sputum and wound drainage fluid culture, and swabs were examined microbiologically for fungi.</p><p><b>RESULTS</b>The incidence of Candida infection in all patients with ANP was 17.9% (14/78) and mortality was 28.6% (4/14). The incidence of prophylactic antibiotic group was 19.4% (12/62) and mortality was 25.0% (3/12). Non prophylactic group was 12.5% (2/16) and 50.0%.</p><p><b>CONCLUSIONS</b>Our data provide evidence for the clinical significance of Candida infection in patients with ANP. The current prophylactic antibiotic treatment can prevent a septic course of the ANP, but might lead to the evolution of Candida infection.</p>
